Article. Published in Chemical Reviews. — 2009. — Vol. 109, №6 — P. 2455–2504Herein, we provide a concise but deep analysis of the protection of amino acids. The review is divided into sections depending on the amino acid funcionalities protected. For each case, methods for the introduction of the protecting groups as well as for their removal are discussed. In eachsection, protecting groups are classified based on the following criteria: (i) the most used in a Boc/Bn strategy; (ii) the most used in a Fmoc/tBu strategy; (iii) decreased order of lability; and (iv) the most recently described, for which, in most cases, their potential has not yet been explored. Inall cases, families of protecting groups are classified together. The compatibility of each protecting group with regard to the others is indicated in the column stability to the removalof, which shows which of the following R-amino-protecting groups (Boc, Fmoc, Z, Trt, Alloc, and pNZ) can be removed without affecting a particular protector.
